Background Information
VAV2 is the second member of the VAV family of guanine nucleotide exchange factors (GEFs) for the Rho family of Ras-related GTPases. As a GEF, it catalyzes the conversion of Rho GTPases from an inactive GDP-bound state to an active GTP-bound state, thereby regulating critical cellular processes such as signal transduction, cytoskeletal remodeling, and cell migration. Unlike its family member VAV1, whose expression is restricted to hematopoietic cells, VAV2 is widely expressed across most tissues. It plays a particularly important role in angiogenesis, where its recruitment by phosphorylated EPHA2 is critical for EFNA1-induced RAC1 GTPase activation and vascular endothelial cell migration and assembly. Structurally, VAV2 contains Dbl homology (DH), Pleckstrin homology (PH), Src homology 2 (SH2), and Src homology 3 (SH3) domains, which are consistent with its function as both a signaling adaptor and an exchange factor.
