Background
Dickkopf-1 (DKK1) acts as a natural inhibitor of Wnt/β-catenin pathway, directly binds to the LRP5/6 receptor and Kremen-1/2, leading to internalization of the receptor complex and dampening of Wnt/β-catenin signaling. Dkk1 is abnormally expressed in tumor cells, and abnormal expression of Dkk1 can inhibit cell proliferation or induce apoptosis through pro-apoptotic factors. In vivo, knocking down of DKK1 in the local site of femoral heads in AONFH rats could result in enhanced bone formation and even restoring the blood supply of the femoral heads.
