Background Information
PTPN23 (Protein Tyrosine Phosphatase Non-receptor Type 23), also known as HD-PTP, is a member of the non-receptor class of protein tyrosine phosphatases that plays a critical role in endosomal trafficking and receptor downregulation. Structurally unique due to its extended C-terminal region containing a Bro1 domain, PTPN23 functions as a core component of the ESCPE-1 (Endosomal SNX-BAR Sorting Complex for Promoting Exit-1) complex and regulates the dephosphorylation of key endosomal proteins, thereby controlling cargo sorting, multivesicular body (MVB) biogenesis, and lysosomal degradation of cell surface receptors such as EGFR. Beyond its role in endocytosis, PTPN23 is involved in ciliogenesis, cell migration, and cytokinesis.
