KE10137 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse Serpin E1/PAI-1 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse Serpin E1/PAI-1. The assay recognizes mouse Serpin E1/PAI-1. An antibody specific for mouse Serpin E1/PAI-1 has been pre-coated onto the microwells. The mouse Serpin E1/PAI-1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse Serpin E1/PAI-1 is added to detect the captured mouse Serpin E1/PAI-1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Serpin E1, also known as plasminogen activator inhibitor 1 (PAI-1), is a protein that belongs to the serpin family of serine protease inhibitors. It is primarily produced by endothelial cells and plays a crucial role in the regulation of fibrinolysis by inhibiting the activity of plasminogen activators, such as tissue plasminogen activator (tPA) and urokinase (uPA). Serpin E1 is involved in a variety of physiological and pathological processes, including fibrinolysis, fibrosis, angiogenesis, wound healing, and the invasion and metastasis of tumor cells.
