Product Information
KE10168 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse HER2/ErbB2 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse HER2/ErbB2. The assay recognizes mouse HER2/ErbB2. An antibody specific for mouse HER2/ErbB2 has been pre-coated onto the microwells. The mouse HER2/ErbB2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse HER2/ErbB2 is added to detect the captured mouse HER2/ErbB2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
HER2/ErbB2 is a critical receptor tyrosine kinase protein belonging to the epidermal growth factor receptor (EGFR) family, which also includes HER1 (EGFR), HER3, and HER4. Unlike other members of the EGFR family, HER2/ErbB2 does not have a known ligand and is often referred to as an "orphan receptor." However, it can form homodimers or heterodimers with other EGFR family members, such as HER1, HER3, and HER4, significantly enhancing its signaling potential. This receptor is involved in the regulation of multiple downstream signaling pathways, including the PI3K-Akt and MAPK pathways, which promote cell survival, proliferation, and migration.
