Product Information
KE10154 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse DR5 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse DR5. The assay recognizes mouse DR5. An antibody specific for mouse DR5 has been pre-coated onto the microwells. The mouse DR5 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse DR5 is added to detect the captured mouse DR5 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
DR5 (Death Receptor 5) is a member of the tumor necrosis factor receptor superfamily, which activates apoptosis signaling pathway mainly through binding to TNF-related apoptosis-inducing ligand (TRAIL). DR5 is expressed in a variety of cell types, including cancer cells and immune cells, and its activation induces apoptosis, thus it is considered as a potential anti-tumor target. In recent years, it has been found that DR5 is not only involved in apoptosis, but also plays an important role in antiviral immunity. For example, DR5 is able to inhibit the replication of human herpesvirus by inducing apoptosis. In addition, activation of DR5 is involved in the integrative stress response (ISR), where the expression of DR5 is increased and ligand-independent activation of the apoptotic pathway under cellular stress conditions. These studies provide new ideas for the development of DR5-based antitumor and antiviral therapies.
