Product Information
KE10131 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse APOE ELISA kit is to be used to detect and quantify protein levels of endogenous mouse APOE. The assay recognizes mouse APOE. An antibody specific for mouse APOE has been pre-coated onto the microwells. The mouse APOE protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse APOE is added to detect the captured mouse APOE protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Apolipoprotein E (APOE) is a core component of plasma lipoproteins and is involved in a variety of biological processes, including plasma lipoprotein metabolism, intracellular cholesterol utilization, cell growth, immune regulation, and neuronal growth and repair. The ε4 allele of the APOE gene is the strongest genetic risk factor for late-onset Alzheimer's disease (AD). Mouse APOE, like ApoE4, contains equivalents of Arg-112 and Glu-255 but lacks the critical Arg-61 equivalent. APOE has heparin-binding activity and binds to heparan sulfate proteoglycans on the cell surface, a property that facilitates cellular capture and receptor-mediated uptake of APOE-containing lipoproteins.
