  • Western Blot (WB) analysis of various lysates using MMP-9 Monoclonal antibody (60600-1-Ig)

    WB analysis using 60600-1-Ig (same clone as 60600-1-PBS)

    Supernatant from non-treated and PMA treated U2OS cells were subjected to SDS PAGE followed by western blot with 60600-1-Ig (MMP-9 antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 60600-1-PBS in a different storage buffer formulation.

    Cytometric bead array of A549 cells supernatant using MP51000-1, MMP-9 Monoclonal Matched Antibody Pair, PBS Only. Capture antibody: 60600-1-PBS. Detection antibody: 60600-2-PBS. A549 cells were cultured in serum-free media overnight followed by PMA stimulation (200 nM, 24 h). 50 μL of the supernatant was analyzed using MP51000-1 (MMP-9 Monoclonal Matched Antibody Pair).     Cytometric bead array of U2OS cells supernatant using MP51000-1, MMP-9 Monoclonal Matched Antibody Pair, PBS Only. Capture antibody: 60600-1-PBS. Detection antibody: 60600-2-PBS. U2OS cells were cultured for 2 days in normal culture media or PMA (200nM, 48h) containing media. 50 μL of the supernatant was analyzed using MP51000-1 (MMP-9 Monoclonal Matched Antibody Pair). Non-related target B2M Monoclonal Matched Antibody Pair (MP50108-1) was served as control.

Background Information

Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, tissue remodeling, and disease processes, such as arthritis or metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. Matrix metalloproteinase 9 (gelatinase B, 92 kDa gelatinase, 92 kDa type IV collagenase) (MMP9, synonyms: GELB, CLG4B) degrades collagens type IV and V. Studies in rhesus monkeys suggest that MMP9 is involved in IL-8-induced mobilization hematopoietic progenitor cells from bone marrow, and murine studies suggest a role in tumor-associated tissue remodeling (PMID:7492685).
