Background Information
Jagunal homolog 1 (JAGN1) is an evolutionarily conserved endoplasmic reticulum (ER) membrane protein essential for neutrophil differentiation and function. First identified in Drosophila for its role in ER organization, human JAGN1 mutations cause severe congenital neutropenia type 5, characterized by recurrent infections due to impaired granulocyte development. JAGN1 regulates glycoprotein folding, ER-Golgi trafficking, and N-glycosylation of proteins critical for neutrophil viability. Loss of JAGN1 leads to ER stress, apoptosis of myeloid progenitors, and defective neutrophil migration. Its indispensable role in innate immunity highlights JAGN1 as a potential therapeutic target for neutropenia and immunodeficiency disorders (PMID: 25129144; 37120535; 33207009; 38739706).
