Background Information
ING1 (Inhibitor of Growth 1) is a tumor suppressor gene located at human chromosome 13q33-34. It encodes multiple protein isoforms generated by alternative splicing and differential promoter usage, including p47ING1a (~47 kDa), p33ING1b (~33 kDa), p24ING1c (~24 kDa), and p27ING1d (~27 kDa) (PMID: 23412501). Among these, p33ING1b is the most abundantly expressed and best-characterized isoform in human cells (PMID: 41617915). All ING1 isoforms share a conserved C-terminal region containing a plant homeodomain (PHD) finger that recognizes the H3K4Me3 histone mark, enabling recruitment of histone deacetylase complexes to regulate chromatin structure and gene transcription. ING1 cooperates with p53 to induce cell cycle arrest and apoptosis, and its dysregulation is implicated in various cancers (PMID: 23255913).
