Background Information
Preparation of Suspension Cell Slides 1. Cell collection: Collect the suspension cell suspension, centrifuge at 1200 rpm for 5 min, then discard the supernatant. 2. Cell washing: Resuspend cells with PBS, centrifuge at 3000 rpm for 1 min, and remove the supernatant. Repeat the washing twice to completely remove residual culture medium. Fixation treatment: Resuspend cells with 4% paraformaldehyde, fix at room temperature for 10-15 min, centrifuge at 3000 rpm for 1 min, then discard the supernatant. 3. Fixative washing: Resuspend cells with PBS, centrifuge at 3000 rpm for 1 min, and discard the supernatant. Repeat the washing twice. 4. Coverslip seeding: Resuspend cells with an appropriate amount of PBS. Add 30 μL of cell suspension per well onto the pre-placed coverslips in a 24-well plate. 5. Drying: Place the plate in an oven at approximately 37 °C and dry for 1.5-2 h. The samples can then be used for subsequent tests such as immunofluorescence assay.
