KE00607 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human ZC3H13 ELISA kit is to be used to detect and quantify protein levels of endogenous human ZC3H13. The assay recognizes human ZC3H13. An antibody specific for human ZC3H13 has been pre-coated onto the microwells. The human ZC3H13 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human ZC3H13 is added to detect the captured human ZC3H13 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

ZC3H13 (zinc finger CCCH-type 13-containing protein) is a protein containing a CCCH-type zinc finger structural domain, which is involved in m6A (N6-methyladenosine) modification, and has an important regulatory role in RNA stability, splicing, and translation. ZC3H13 is aberrantly expressed in a variety of cancers, e.g. in hepatocellular carcinoma (HCC), where its down-regulated expression correlates with poor prognosis, while overexpression inhibits the tumor cell proliferation and invasion. In colorectal cancer, ZC3H13 inhibits tumor cell proliferation and invasion by suppressing the Ras-ERK signaling pathway. In addition, ZC3H13 plays an important role in cardiovascular diseases, such as in septic cardiomyopathy (SCM), where its expression is up-regulated, and silencing of ZC3H13 increases the expression of proteins related to cell proliferation and iron death, and reduces apoptosis and reactive oxygen species (ROS) accumulation. ZC3H13 has also been associated with neurological disorders, and its aberrant expression may be related to neurodegenerative diseases.
