Product Information
KE00538 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human USP8 ELISA kit is to be used to detect and quantify protein levels of endogenous human USP8. The assay recognizes human USP8. An antibody specific for human USP8 has been pre-coated onto the microwells. The human USP8 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human USP8 is added to detect the captured human USP8 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
USP8 is a member of the USP family of deubiquitinases, it is a cysteine protease that plays a crucial role in regulating protein ubiquitination and degradation. The activity of USP8 is regulated by phosphorylation and protein-protein interactions. Mutations in the USP8 gene, particularly in the 14-3-3 binding motif, have been implicated in corticotroph adenomas, a type of pituitary tumor.
