KE00775 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TSLP ELISA kit is to be used to detect and quantify protein levels of endogenous human TSLP. The assay recognizes human TSLP. An antibody specific for human TSLP has been pre-coated onto the microwells. The human TSLP protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TSLP is added to detect the captured human TSLP protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Thymic stromal lymphopoietin (TSLP) is a multifunctional cytokine primarily produced by epithelial cells in response to tissue damage, infection, or allergen exposure. It plays a significant role in immune regulation, inflammation, and cancer. TSLP acts through its receptor complex, composed of TSLP receptor (TSLPR) and interleukin-7 receptor alpha chain (IL-7Rα), to activate downstream signaling pathways, including JAK-STAT, PI3K-AKT, and MAPK. TSLP is involved in various physiological and pathological conditions, including asthma, allergic diseases, and chronic inflammatory diseases. It promotes type 2 immune responses by inducing the release of T cell-attracting chemokines from monocytes and enhancing the maturation of CD11c(+) dendritic cells. Additionally, TSLP plays a role in tissue homeostasis, host defense, and microbiome regulation.
