KE00573 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TSG101 ELISA kit is to be used to detect and quantify protein levels of endogenous human TSG101. The assay recognizes human TSG101. An antibody specific for human TSG101 has been pre-coated onto the microwells. The human TSG101 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TSG101 is added to detect the captured human TSG101 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
TSG101 (tumor susceptibility gene 101 protein) is a multi-structural domain protein that is widely expressed in a variety of cells and is involved in a variety of intracellular processes. It plays a key role in endoplasmic reticulum sorting, membrane receptor degradation, and the final stages of cytokinesis. TSG101 was initially defined as a tumor suppressor gene, but in some epithelial tumor cells, it exhibits tumor-promoting effects. In addition, TSG101 is involved in exosome secretion, positive regulation of ubiquitin-dependent endocytosis, and viral outgrowth. It also plays an important role in cell cycle regulation, cell survival, and maintenance of genome stability.
