KE00550 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TIM3 ELISA kit is to be used to detect and quantify protein levels of endogenous human TIM3. The assay recognizes human TIM3. An antibody specific for human TIM3 has been pre-coated onto the microwells. The human TIM3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TIM3 is added to detect the captured human TIM3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
TIM3 (T-cell immunoglobulin and mucin-domain containing molecule 3), also known as HAVCR2, is an immune checkpoint receptor expressed on various immune cells, including T cells, dendritic cells, and macrophages. It plays a crucial role in regulating immune responses by mediating inhibitory signals contributing to immune tolerance and exhaustion. TIM3 is highly expressed on tumor-infiltrating lymphocytes (TILs) and is associated with poor prognosis in several cancers, including colorectal cancer. In cancer, soluble TIM3 may contribute to immune evasion by binding to galectin-9 and other ligands, thereby inhibiting the function of immune cells.
