KE00524 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TBX21/T-bet ELISA kit is to be used to detect and quantify protein levels of endogenous human TBX21/T-bet. The assay recognizes human TBX21/T-bet. An antibody specific for human TBX21/T-bet has been pre-coated onto the microwells. The human TBX21/T-bet protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TBX21/T-bet is added to detect the captured human TBX21/T-bet protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
TBX21, also known as T-bet, is a key T-box transcription factor that plays a critical role in the regulation of immune responses and the development of Th1 cells. The gene is a member of the phylogenetically conserved T-box family, which shares a common DNA-binding domain. T-bet is predominantly expressed in Th1 cells and natural killer (NK) cells, where it controls the expression of the hallmark Th1 cytokine, interferon-gamma (IFN-γ). This transcription factor is essential for initiating Th1 lineage development from naïve T precursor cells.
