Product Information
KE00832 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TBC1D5 ELISA kit is to be used to detect and quantify protein levels of endogenous human TBC1D5. The assay recognizes human TBC1D5. An antibody specific for human TBC1D5 has been pre-coated onto the microwells. The human TBC1D5 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TBC1D5 is added to detect the captured human TBC1D5 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
TBC1D5 (TBC1 domain family member 5), a TBC family protein, is a critical retromer component, mediating the reverse transport of target molecules from endosomes to the Golgi apparatus. TBC1D5 and retromer complex can control the activity, localization, and mobility of RAB7, further regulating the biological process of RAB7 involved, especially those correlated with autophagy. The TBC1D5 molecule contains two LC3-acting elements (LC3-interacting region, Lir), and both are involved in directly binding TBC1D5 and MAP1LC3, further participating in forming autophagosomes from the initial stage. TBC1D5 can also regulate the initiation of mitophagy.
