Product Information
KE00539 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Siglec-8 ELISA kit is to be used to detect and quantify protein levels of endogenous human Siglec-8. The assay recognizes human Siglec-8. An antibody specific for human Siglec-8 has been pre-coated onto the microwells. The human Siglec-8 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Siglec-8 is added to detect the captured human Siglec-8 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Sialic acid-binding Ig-like lectin 8 (Siglec-8) is a novel sialic acid-binding immunoglobulin-like lectin that is uniquely expressed by human eosinophils, mast cells, as well as basophils. It has homology with other sialoadhesin family members (CD33 and siglec-5) and belongs to a subgroup of the Ig superfamily. Siglec-8 also exists in a soluble form in human serum (sSiglec-8), and reports suggested that sSiglec-8 might be used as a biomarker for various eosinophil-associated diseases.
