KE00671 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human S100A9 ELISA kit is to be used to detect and quantify protein levels of endogenous human S100A9. The assay recognizes human S100A9. An antibody specific for human S100A9 has been pre-coated onto the microwells. The human S100A9 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human S100A9 is added to detect the captured human S100A9 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
S100A9 (S100 calcium-binding protein A9) is also named as Calgranulin-B (CAGB), CFAG and MRP14. S100A9 is a member of the S100 protein family. It is a calcium‐binding protein that was first identified as a proinflammatory mediator released by bone marrow‐derived cells in response to cell damage, infection, or inflammation. S100A9 is also involved in carcinogenesis and tumor progression and inhibits the tumor immune response. S100A9 might activate the reactive oxygen species (ROS)‐dependent signaling pathways and protect against cell death. S100A9 acts as a scaffold recruiting ubiquitin specific peptidase 10 and phosphoglycerate mutase family member 5 (PGAM5) to form a tripolymer, causing the deubiquitination and stabilization of PGAM5, leading to mitochondrial fission and reactive oxygen species production, thereby promoting the growth and metastasis of HCC . The alarmins S100A8 and S100A9 are abundantly stored in neutrophils as the stable heterodimer S100A8/A9, also known as calprotectin, and are rapidly released in response to inflammatory stimuli and act as damage-associated molecular patterns.5 Neutrophils are the most important source of extracellular S100A8/A9.
