KE00636 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human RNF40 ELISA kit is to be used to detect and quantify protein levels of endogenous human RNF40. The assay recognizes human RNF40. An antibody specific for human RNF40 has been pre-coated onto the microwells. The human RNF40 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human RNF40 is added to detect the captured human RNF40 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
RNF40 (Ring Finger Protein 40) is an E3 ubiquitin ligase, which is involved in the monoubiquitination of histone H2B at lysine 120 (H2Bub1), a modification that is crucial for transcriptional elongation and DNA repair. RNF20, RNF40 and H2Bub1 were also found to correlate with poor prognosis in hepatocellular carcinoma and to support the tumorigenic properties of prostate cancer and MLL-rearranged lymphoblastic leukemia.
