Product Information
KE00950 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human RGS12 ELISA kit is to be used to detect and quantify protein levels of endogenous human RGS12. The assay recognizes human RGS12. An antibody specific for human RGS12 has been pre-coated onto the microwells. The human RGS12 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human RGS12 is added to detect the captured human RGS12 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
RGS12 (Regulator of G-protein Signaling 12) belongs to the superfamily of RGS proteins defined by a conserved RGS domain that canonically binds and deactivates heterotrimeric G-proteins. It negatively regulates G-protein-coupled receptor (GPCR) signaling by accelerating GTP hydrolysis on Gα subunits. Unlike typical RGS proteins, RGS12 uniquely integrates multiple signaling domains, including a PDZ domain, PTB domain, and Ras-binding domain (RBD), enabling it to orchestrate cross-talk between GPCRs, receptor tyrosine kinases (RTKs), and mitogen-activated protein kinase (MAPK) pathways. It plays critical roles in neuronal development, osteoclast differentiation, and immune response by modulating calcium channels, transcription factors, and cell cycle progression.
