Product Information
KE00850 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human RAD50 ELISA kit is to be used to detect and quantify protein levels of endogenous human RAD50. The assay recognizes human RAD50. An antibody specific for human RAD50 has been pre-coated onto the microwells. The human RAD50 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human RAD50 is added to detect the captured human RAD50 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Rad50, an ATPase, binds to Mre11 and Nbs1 to form a repair complex that is transported to the nucleus in response to DNA double-strand breaks. Rad50 is a component of MRN (Mre11-Rad50-Nbs1), which participates in DNA double-strand break repair and DNA-damage checkpoint activation. Rad50 promoted proliferation/invasion and induced EMT of ovarian cancer cells, whereas knockdown of Rad50 led to decreased aggressive behaviors . Frequent lack of RAD50 was found in endometrial and low-grade epithelial ovarian cancer, whereas Rad50 was highly expressed in melanoma and gastric cancer.
