KE01047 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PTPN12 ELISA kit is to be used to detect and quantify protein levels of endogenous human PTPN12. The assay recognizes human PTPN12. An antibody specific for human PTPN12 has been pre-coated onto the microwells. The human PTPN12 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PTPN12 is added to detect the captured human PTPN12 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
PTPN12 (Tyrosine-protein phosphatase non-receptor type 12), also referred to as PTP-PEST (PTP-proline, glutamic acid, serine, and threonine-rich), is a cytoplasmic PTP expressed in a wide spectrum of cell types. PTPN12 has been identified as a potent tumor suppressor in human cancers and a critical regulator of cell adhesion and migration. PTPN12 is phosphorylated on serine and threonine residues and is rich in proline (P), glutamate (E), serine (S), and threonine (T) residues.
