Product Information

KE00692 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PON3 ELISA kit is to be used to detect and quantify protein levels of endogenous human PON3. The assay recognizes human PON3. An antibody specific for human PON3 has been pre-coated onto the microwells. The human PON3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PON3 is added to detect the captured human PON3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

PON3 (Paraoxonase 3) is a member of the PON protein family, primarily synthesized by the liver and secreted into the bloodstream, where it binds to high-density lipoprotein (HDL). It has antioxidant functions and can rapidly hydrolyze lactones, including statin precursors (such as lovastatin), and inhibit the oxidation of low-density lipoprotein (LDL), thereby slowing the development of atherosclerosis. Additionally, PON3 exerts antioxidant effects within cells, reducing oxidative stress and protecting cells from oxidative damage. In cancer tissues, the expression of PON3 is upregulated, and it functions on the mitochondrial membrane of cancer cells, protecting them from mitochondrial superoxide-mediated cell death. These characteristics of PON3 endow it with potential protective roles in pathological processes such as cardiovascular diseases, metabolic syndrome, neurodegenerative diseases, and cancer.
