Product Information
KE00543 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PON1 ELISA kit is to be used to detect and quantify protein levels of endogenous human PON1. The assay recognizes human PON1. An antibody specific for human PON1 has been pre-coated onto the microwells. The human PON1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PON1 is added to detect the captured human PON1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
PON1 (Paraoxonase 1) is a serum enzyme primarily synthesized in the liver and associated with high-density lipoproteins (HDL). It is capable of hydrolyzing a broad spectrum of organophosphate substrates and several aromatic carboxylic acid esters. PON1 plays a crucial role in detoxifying organophosphate compounds and has antioxidant properties that protect against oxidative stress. PON1 may mediate enzymatic protection of low-density lipoproteins against oxidative modification and the consequent series of events leading to atheroma formation.
