KE00994 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PDE5A ELISA kit is to be used to detect and quantify protein levels of endogenous human PDE5A. The assay recognizes human PDE5A. An antibody specific for human PDE5A has been pre-coated onto the microwells. The human PDE5A protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PDE5A is added to detect the captured human PDE5A protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
PDE5A (cGMP-binding cGMP-specific phosphodiesterase) plays a role in signal transduction by regulating the intracellular concentration of cyclic nucleotides. This phosphodiesterase catalyzes the specific hydrolysis of cGMP to 5'-GMP. Through alternative splicing, the PDE5A gene produces three proteins (PDE5A1, PDE5A2, and PDE5A3) that differ in their N termini and range in mass from 89 to 105 kDa. A second 70 kDa band was consistently observed in heart tissue that either reflected a splice variant or a proteolytic fragment of PDE5A.
