Product Information

KE00341 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PARK7/DJ-1 ELISA kit is to be used to detect and quantify protein levels of endogenous human PARK7/DJ-1. The assay recognizes human PARK7/DJ-1. An antibody specific for human PARK7/DJ-1 has been pre-coated onto the microwells. The human PARK7/DJ-1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PARK7/DJ-1 is added to detect the captured human PARK7/DJ-1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

PARK7, also named as DJ1, belongs to the peptidase C56 family. It protects cells against oxidative stress and cell death. PARK7 plays a role in regulating expression or stability of the mitochondrial uncoupling proteins SLC25A14 and SLC25A27 in dopaminergic neurons of the substantia nigra pars compacta and attenuates the oxidative stress induced by calcium entry into the neurons via L-type channels during pacemaking. It eliminates hydrogen peroxide and protects cells against hydrogen peroxide-induced cell death. PARK7 has cell-growth promoting activity and transforming activity. It may function as a redox-sensitive chaperone. It's precursor undergoes a cleavage of a C-terminal peptide and subsequent activation of protease activity in response to oxidative stress.
