Product Information
KE00570 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human OGT ELISA kit is to be used to detect and quantify protein levels of endogenous human OGT. The assay recognizes human OGT. An antibody specific for human OGT has been pre-coated onto the microwells. The human OGT protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human OGT is added to detect the captured human OGT protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
O-linked N-acetylglucosamine transferase (OGT) catalyzes the attachment of N-acetylglucosamine (GlcNAc) monosaccharides to the hydroxyl group of serine or threonine residues of numerous nuclear and cytoplasmic proteins and may play important roles in a large number of diverse intracellular processes ranging from translational control, transcription, transcriptional repression, INS resistance and regulation of the cell cycle. It exists as a heterotrimeric complex with two 110 kDa and one 70 kDa subunits. Recent studies have shown that O-GlcNAcylation plays essential roles in cancer formation and progression. O-GlcNAcylation as well as OGT expression was found to be significantly elevated in the cancer tissues.
