Product Information
KE00975 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human ND4 ELISA kit is to be used to detect and quantify protein levels of endogenous human ND4. The assay recognizes human ND4. An antibody specific for human ND4 has been pre-coated onto the microwells. The human ND4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human ND4 is added to detect the captured human ND4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
MTND4 is one of the core mitochondrial-encoded subunits of the mitochondrial membrane respiratory chain NADH dehydrogenase (Complex I). MTND4 gene codes for the NADH-ubiquinone oxidoreductase chain 4 (ND4) protein . The ND4 protein is located in the mitochondrial inner membrane and is the largest of the five complexes of the electron transport chain . Complex I is the entry point of electrons to the OXPHOS system, transferring electrons from NADH (reduced nicotinamide adenine dinucleotide) to ubiquinone; the electron transference is coupled to the proton pumping inside the mitochondrial intermembrane space (IMS) to form, in part, the proton gradient used to produce ATP. Interestingly, MTND4 is essential for the assembly and proper activity of the Complex I.
