KE00414 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse/rat PLEC ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse/rat PLEC. The assay recognizes human/mouse/rat PLEC. An antibody specific for human/mouse/rat PLEC has been pre-coated onto the microwells. The human/mouse/rat PLEC protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse/rat PLEC is added to detect the captured human/mouse/rat PLEC protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
PLEC (Plectin) is a large protein normally expressed in various tissues including skin, muscle, and brain. It binds to many cytoskeletal proteins including microtubules and intermediate filaments and is involved in the establishment and dynamic modulation of the cytoskeletal network. In normal cells, it plays a crucial role in cytoskeleton network organization. In keratinocytes, plectin is concentrated at the basal surface, where it links intermediate filaments to the cytoplasmic domain of transmembrane glycoproteins such as integrin beta-4.
