KE01030 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse/rat PCNA ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse/rat PCNA. The assay recognizes human/mouse/rat PCNA. An antibody specific for human/mouse/rat PCNA has been pre-coated onto the microwells. The human/mouse/rat PCNA protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse/rat PCNA is added to detect the captured human/mouse/rat PCNA protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Proliferating Cell Nuclear Antigen, commonly known as PCNA, is a protein that acts as a processivity factor for DNA polymerase δ in eukaryotic cells. This protein is an auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase's processibility during elongation of the leading strand. PCNA induces a robust stimulatory effect on the 3'-5' exonuclease and 3'-phosphodiesterase, but not apurinic-apyrimidinic (AP) endonuclease, APEX2 activities. It has to be loaded onto DNA to stimulate APEX2. PCNA, a protein synthesized in early G1 and S phases of the cell cycle, functions in cell cycle progression, DNA replication, and DNA repair. In the early S phase, PCNA exhibits granular distribution and is absent from the nucleoli; however, in the late S phase, it relocates to the nucleoli.
