Product Information
KE00891 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse/rat HCN4 ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse/rat HCN4. The assay recognizes human/mouse/rat HCN4. An antibody specific for human/mouse/rat HCN4 has been pre-coated onto the microwells. The human/mouse/rat HCN4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse/rat HCN4 is added to detect the captured human/mouse/rat HCN4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
HCN4 (Hyperpolarization-activated Cyclic Nucleotide-gated Channel 4) belongs to the potassium channel HCN family. It is a hyperpolarization-activated ion channel with very slow activation and inactivation, exhibiting weak selectivity for potassium over sodium ions. HCN4 may contribute to the native pacemaker currents in the heart (If) and in neurons (Ih). It is regulated by cyclic nucleotides such as cAMP, which modulate channel activity by shifting the voltage dependence of activation and altering the rate of channel opening and closing. HCN4 may mediate responses to sour stimuli. Defects in HCN4 are a cause of sick sinus syndrome type 2 (SSS2).
