Product Information
KE00639 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse/rat HCN2 ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse/rat HCN2. The assay recognizes human/mouse/rat HCN2. An antibody specific for human/mouse/rat HCN2 has been pre-coated onto the microwells. The human/mouse/rat HCN2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse/rat HCN2 is added to detect the captured human/mouse/rat HCN2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels are a group of voltage-gated ion channels in which the threshold voltage is modulated by the metabolic state of the cell (levels of cyclic nucleotides, like cAMP), thus allowing them to regulate cellular voltage in a context-specific manner. They open at hyperpolarized (negative) Vmem, giving rise to currents that are a mix of sodium and potassium fluxes. Among the four HCN isoforms (HCN1-4), HCN2 channels show an especially high sensitivity to cAMP.
