Product Information
KE00840 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse/rat GSR ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse/rat GSR. The assay recognizes human/mouse/rat GSR. An antibody specific for human/mouse/rat GSR has been pre-coated onto the microwells. The human/mouse/rat GSR protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse/rat GSR is added to detect the captured human/mouse/rat GSR protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
GSR (glutathione-disulfide reductase) is also named as glutathione reductase (GR), GLUR, GRD1, which is an enzyme that catalyzes the reduction of glutathione disulfide (GSSG) to the sulfhydryl form of glutathione (GSH), which is a critical molecule in resisting oxidative stress and maintaining the reducing environment of the cell. Glutathione plays a key role in maintaining proper function and preventing oxidative stress by acting as a scavenger for hydroxyl radicals, singlet oxygen, and various electrophiles. Reduced glutathione reduces the oxidized form of the enzyme glutathione peroxidase, which in turn reduces H2O2, a dangerously reactive species within the cell. In addition, GSH plays a key role in the metabolism and clearance of xenobiotics, acts as a cofactor in certain detoxifying enzymes, and regenerates antioxidants such as vitamins C and E to their reactive forms. The ratio of GSSG/GSH present in the cell is a key factor in maintaining the oxidative cell balance, where it is critical that a cell maintains high levels of the reduced glutathione and a low level of oxidized glutathione disulfide. This narrow balance is "buffered" by GSR, which catalyzes the reduction of GSSG to GSH.
