KE00966 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse/rat ATOH1 ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse/rat ATOH1. The assay recognizes human/mouse/rat ATOH1. An antibody specific for human/mouse/rat ATOH1 has been pre-coated onto the microwells. The human/mouse/rat ATOH1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse/rat ATOH1 is added to detect the captured human/mouse/rat ATOH1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
ATOH1, also named as Protein atonal homolog 1 or BHLHA14, is a 364 amino acid protein, which contains 1 bHLH (basic helix-loop-helix) domain. ATOH1,as a transcriptional regulator in the nucleus activates E box-dependent transcription in collaboration with TCF3/E47, but the activity is completely antagonized by the negative regulator of neurogenesis HES1. ATOH1 is involved in differentiation of neurons, secretory cells in the gut, and mechanoreceptors including auditory hair cells. ATOH1 impact in cancer is context dependent, described as a tumor suppressor in colorectal cancer and an oncogene in medulloblastoma.
