KE00664 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/mouse PPIF ELISA kit is to be used to detect and quantify protein levels of endogenous human/mouse PPIF. The assay recognizes human/mouse PPIF. An antibody specific for human/mouse PPIF has been pre-coated onto the microwells. The human/mouse PPIF protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/mouse PPIF is added to detect the captured human/mouse PPIF protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
PPIF, also known as Cyclophilin D (CypD), CYP3, Rotamase F, is a highly conserved peptidyl-prolyl cis-trans isomerase (PPIase) that plays an important role in mitochondrial biology. PPIF has been thought to regulate the opening of the permeability transition pore (PTP), a large conductance pore whose opening depolarizes the inner mitochondrial membrane (IMM) to decrease ATP and increase reactive oxygen species production. PPIF is a key mitochondrial target for Aβ-induced mitochondrial and synaptic dysfunction. Blockade of PPIF may be a potential therapeutic approach for halting Alzheimer's disease.
