Product Information
KE00826 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human MXI1 ELISA kit is to be used to detect and quantify protein levels of endogenous human MXI1. The assay recognizes human MXI1. An antibody specific for human MXI1 has been pre-coated onto the microwells. The human MXI1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human MXI1 is added to detect the captured human MXI1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
MXI1, also known as MAX Interactor 1, is a transcription factor containing a basic helix-loop-helix leucine zipper (bHLHzip) domain and belongs to the Myc-Max-Mad transcriptional network. It is a widely expressed transcriptional repressor that heterodimerizes with Max and blocks Max-Myc gene activation. MXI1 is involved in the homeostasis of differentiated organ systems and acts as a tumor suppressor in vivo. It has been implicated in several diseases, including prostate cancer and neurofibrosarcoma. In lung cancer, MXI1 expression is often low, and its poor expression is associated with poor prognosis. MXI1 is also involved in other biological processes. For example, in mesangial proliferative glomerulonephritis, MXI1 expression changes during the disease process, influencing mesangial cell proliferation. Additionally, MXI1 has been shown to be directly regulated by the HIF-1α protein in neuroblastoma and breast cancer cells.
