Product Information
KE00522 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human MTHFR ELISA kit is to be used to detect and quantify protein levels of endogenous human MTHFR. The assay recognizes human MTHFR. An antibody specific for human MTHFR has been pre-coated onto the microwells. The human MTHFR protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human MTHFR is added to detect the captured human MTHFR protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
5,10-Methylenetetrahydrofolate reductase (MTHFR) is a crucial enzyme in the folate metabolic pathway with a key role in generating methyl groups. MTHFR reduces 5,10-methylenetetrahydrofolate (5,10-methyleneTHF) to 5-methylTHF, the primary carbon donor for methionine production from homocysteine. In human and mouse studies, mutations and polymorphisms in MTHFR are associated with impaired spermatogenesis and male infertility. It has been reported that MTHFR is highly phosphorylated under unperturbed conditions and T34 is the priming phosphorylation site.
