Product Information
KE00438 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human MNT ELISA kit is to be used to detect and quantify protein levels of endogenous human MNT. The assay recognizes human MNT. An antibody specific for human MNT has been pre-coated onto the microwells. The human MNT protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human MNT is added to detect the captured human MNT protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
MNT (MAX network transcriptional repressor ), also known as MAD6, ROX, bHLHd3 (class D basic helix-loop-helix protein 3) or MXD6, is a 582 amino acid nuclear protein. Mnt contains one basic helix-loop-helix (bHLH) domain, and Mnt acts primarily as a transcriptional repressor. It binds to the Max(Myc-associated factor X) protein and inhibits the expression of target genes that are typically activated by Myc. This repressive action is crucial in maintaining the balance between cell growth and apoptosis, preventing uncontrolled cell proliferation. It predicts that antagonism of Myc by Mxd-family proteins and Mnt plays significant roles in regulating the various activities of Myc, including its role in tumorigenesis.
