Product Information
KE00483 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human MCM2 ELISA kit is to be used to detect and quantify protein levels of endogenous human MCM2. The assay recognizes human MCM2. An antibody specific for human MCM2 has been pre-coated onto the microwells. The human MCM2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human MCM2 is added to detect the captured human MCM2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
MCM2 (microchromosome maintenance protein 2) is an important member of the family of microchromosome maintenance proteins (MCM family of proteins) involved in the initiation and elongation of DNA replication in eukaryotic cells. MCM2, together with MCM3-MCM7, forms the hexameric MCM2-7 complex, which serves as a replicative deconjugating enzyme that utilizes the energy of ATP hydrolysis to drive DNA deconjugation, ensuring that DNA is replicated only once per cell cycle. MCM2 is loaded onto chromatin during the G1 phase of the cell cycle and is a marker for S-phase cells. Its expression level is closely related to cell proliferation and is up-regulated in a variety of tumors (e.g. breast cancer, colorectal cancer, lung cancer, etc.), and can be used as a marker for precancerous lesions and tumor progression.
