KE00953 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human MAFF ELISA kit is to be used to detect and quantify protein levels of endogenous human MAFF. The assay recognizes human MAFF. An antibody specific for human MAFF has been pre-coated onto the microwells. The human MAFF protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human MAFF is added to detect the captured human MAFF protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
MAFF is a member of the small Maf protein family (MafF, MafG, MafK) within the basic region leucine zipper (bZIP) transcription factor superfamily. It can form homodimers or heterodimers with other bZIP proteins (e.g., Nrf2, Bach1) to regulate gene expression by binding to antioxidant response elements (AREs) or Maf recognition elements (MAREs). MAFF plays critical roles in cellular defense against oxidative stress, erythrocyte differentiation, and neuronal development. It is highly expressed in specific tissues, including the liver, kidney, and hematopoietic cells.
