KE00574 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human LIN28A ELISA kit is to be used to detect and quantify protein levels of endogenous human LIN28A. The assay recognizes human LIN28A. An antibody specific for human LIN28A has been pre-coated onto the microwells. The human LIN28A protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human LIN28A is added to detect the captured human LIN28A protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
LIN28 is one of the four key human factors (OCT4, SOX2, NANOG and LIN28) used to reprogram human fibroblasts to an embryonic stem (ES) cell-like state known as the induced pluripotent stem (Ips) cell. LIN28 is a marker of undifferentiated human embryonic stem cells and a cytoplasmic mRNA-binding protein that binds to and enhances the translation of the IGF2 mRNA. LIN28 has also been shown to bind to the let-7 pre-miRNA and block production of the mature let-7 microRNA in mouse embryonic stem cells.
