KE00575 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human IRAK3 ELISA kit is to be used to detect and quantify protein levels of endogenous human IRAK3. The assay recognizes human IRAK3. An antibody specific for human IRAK3 has been pre-coated onto the microwells. The human IRAK3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human IRAK3 is added to detect the captured human IRAK3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
IRAK3 (Interleukin-1 receptor-associated kinase 3) is a cytoplasmic homeostatic mediator of inflammatory responses and is potentially useful as a prognostic marker in inflammation. IRAK3 is associated with many inflammatory diseases, including sepsis, and is required in endotoxin tolerance to maintain homeostasis of inflammation. IRAK3 is expressed in macrophages and pancreatic acinar cells where it restrains NFκB activation.
