Product Information
KE00935 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human IMPDH1 ELISA kit is to be used to detect and quantify protein levels of endogenous human IMPDH1. The assay recognizes human IMPDH1. An antibody specific for human IMPDH1 has been pre-coated onto the microwells. The human IMPDH1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human IMPDH1 is added to detect the captured human IMPDH1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
IMPDH1 (inosine monophosphate dehydrogenase 1) is a key enzyme involved in the de novo synthesis pathway of purine nucleotides. It catalyzes the conversion of inosine monophosphate (IMP) to xanthosine monophosphate (XMP), which is a rate-limiting step in the synthesis of guanosine monophosphate (GMP). IMPDH1 exists as a tetramer within cells and is crucial for maintaining the balance of the intracellular guanine nucleotide pool. This balance affects processes related to cell proliferation, including DNA and RNA synthesis, signal transduction, and energy transfer. Moreover, abnormal expression of IMPDH1 is associated with various diseases, including cancer and autoimmune diseases. In the retina, mutations in IMPDH1 are linked to autosomal dominant retinitis pigmentosa (adRP) type RP10. These mutations primarily affect the allosteric regulation of the enzyme rather than its catalytic activity, leading to disruptions in guanine nucleotide metabolism in the retina.
