KE00782 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human IGFBP3 ELISA kit is to be used to detect and quantify protein levels of endogenous human IGFBP3. The assay recognizes human IGFBP3. An antibody specific for human IGFBP3 has been pre-coated onto the microwells. The human IGFBP3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human IGFBP3 is added to detect the captured human IGFBP3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Insulin-like growth factor binding protein-3 (IGFBP-3) contains a 27 amino acid putative signal sequence followed by a mature protein of 264 amino acids with 18 cysteine residues clustered near the N- and C-terminus. Accordingly, expression of the cloned IGFBP-3 cDNA in mammalian tissue culture cells results in secretion of the protein into the culture medium. IGFBP-3 shares high homology (33% amino acid identity) including conservation of all 18 cysteine residues with a smaller human IGF-binding protein (BP-28) identified in amniotic fluid. IGFBP-3 has one or more glycosylation sites with a protein core of ∼30 kDa. Western blots revealed that the 39-45 kDa IGFBP-3 fragment is a glycoprotein.
