Product Information
KE00634 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human HNF4A ELISA kit is to be used to detect and quantify protein levels of endogenous human HNF4A. The assay recognizes human HNF4A. An antibody specific for human HNF4A has been pre-coated onto the microwells. The human HNF4A protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human HNF4A is added to detect the captured human HNF4A protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
HNF4A (Hepatocyte nuclear factor 4 α) is highly conserved between humans and mice. Using conditional knockouts, epithelial HNF4A is indispensable for protecting against acute chemical-induced colitis and spontaneous colonic inflammation in aged animals. Moreover, reduced expression of HNF4A has been reported in liver fibrosis, and viral vector-mediated HNF4A restoration has previously been demonstrated to reverse features of chronic liver injury. Therefore, HNF4A has been suggested as a therapeutic target for treating liver fibrosis.
