Product Information

KE00717 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human HIRA ELISA kit is to be used to detect and quantify protein levels of endogenous human HIRA. The assay recognizes human HIRA. An antibody specific for human HIRA has been pre-coated onto the microwells. The human HIRA protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human HIRA is added to detect the captured human HIRA protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

HIRA (Histone Regulator A) is a key histone chaperone involved in the deposition of the histone variant H3.3 into chromatin. The histone chaperone HIRA complex, composed of HIRA, Cabin1 and UBN1, is an evolutionarily conserved histone chaperone, which has been widely reported as a critical chromatin regulator in senescent cells. Moreover, other histone chaperones assist HIRA in performing multiple activities: 1. DNA damage repair, 2. H3.3 enrichment regulation, 3. myogenic progression, etc.
