KE00860 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human GPR56 ELISA kit is to be used to detect and quantify protein levels of endogenous human GPR56. The assay recognizes human GPR56. An antibody specific for human GPR56 has been pre-coated onto the microwells. The human GPR56 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human GPR56 is added to detect the captured human GPR56 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

GPR56 (G protein-coupled receptor 56) is an adhesion G protein-coupled receptor (aGPCR) that belongs to the G protein-coupled receptor family. It regulates various cellular functions through interactions between its N-terminal GAIN domain and exogenous ligands. GPR56 plays a critical role in nervous system development, particularly in the development and myelination of oligodendrocytes. Additionally, it is involved in interactions with the extracellular matrix, binding to molecules such as collagen III and tissue transglutaminase (TG2) to regulate cell adhesion and migration. GPR56 is abnormally expressed in a variety of diseases, including liver fibrosis, depression, and multiple tumors. In liver fibrosis, GPR56 alleviates hepatocyte pyroptosis by inhibiting the NF-κB/NLRP3 pathway, thereby blocking the activation of hepatic stellate cells. In the field of oncology, GPR56 is considered a potential tumor biomarker and therapeutic target.
