Product Information

KE00571 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human GPNMB ELISA kit is to be used to detect and quantify protein levels of endogenous human GPNMB. The assay recognizes human GPNMB. An antibody specific for human GPNMB has been pre-coated onto the microwells. The human GPNMB protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human GPNMB is added to detect the captured human GPNMB protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

GPNMB (Glycoprotein non-metastatic melanoma protein B), also known as osteoactivin, is a type I transmembrane glycoprotein that plays a role in various physiological processes including cell differentiation, migration, inflammation, metabolism, and neuroprotection. It is expressed in multiple tissues such as bone, kidney, and skeletal muscle, and is implicated in cell processes like tissue regeneration and tumor progression. GPNMB has also been identified within motor neurons and radial glia, most abundantly in microglia cells, suggesting its role in inflammatory processes in the central nervous system. Its role in inflammation and tissue regeneration makes it a potential therapeutic target in diseases like atherosclerosis.
